Can't access Windows Task Manager, although I'm admin?

My computer has been slowing down often lately, with these sounds coming from the HD like something was working in the background… it's happened before, but I was able to just close the rogue processes from the Task Manager, and everything would go back to normal. Now when I try that (press Ctrl-Alt-Del), nothing happens! I don't know what's going on, I don't want to restart every few hours to deal with this…

Well, the easy solution would be to try Ctrl + Shift + Esc instead of the tried and true Ctrl + Alt + Del, maybe a key is just malfunctioning.
But more importantly, you need to deal with the problem at the source, not just use Task Manager for temporary relief. Check for spyware and viruses – if you don't have any tools that do that, search for free scans online.
